◆ Compared with conventional blood DNA sample collection, urine DNA sample collection is a new non-invasive and painless method for human DNA sample collection. The urine DNA sample preservation tube can preserve not only the genome in urine but also cell-free circulating DNA (cfDNA) in urine. Studies have demonstrated that part of plasma cfDNA originates from tumors, fetuses and transplanted organs, and plasma cell-free DNA can pass through the kidneys into urine, which brings great application value for non-invasive diagnosis.
◆ After urine sample collection, the urine DNA sample preservation tube can stably preserve DNA at 6–35°C for up to 7 days. Composed of EDTA anticoagulant and a special protective agent, it can effectively inhibit nucleases in urine, prevent the release of genomic DNA from exfoliated cells in urine, and also preserve blood cells in urine. Featuring convenient collection, non-invasiveness, and storage and transportation at room temperature, it greatly facilitates the transportation and storage of urine samples. This product is suitable for research related to non-invasive prenatal screening and early diagnosis of certain diseases.
◆ The urine DNA sample preservation tube is a negative-pressure tube made of PET material, with a built-in fixed volume of urine DNA sample preservation solution, used for the collection and preservation of urine samples from urine collection cups.
◆ Precautions for Urine Collection
1. It is recommended to collect urine volume sufficient for 2 urine DNA sample preservation tubes. Excessive or insufficient urine volume will lead to an inappropriate urine/protective agent ratio and fail to achieve optimal test results.
2. Discontinue use if precipitation or insoluble substances are found in the product before use.
3. First-morning urine is preferred. Invert the preservation tube gently up and down 10 times immediately after urine collection. Insufficient or excessive mixing may affect the test results.
◆ Cell-Free DNA Extraction Method
1. Urine separation: Centrifuge the urine DNA sample preservation tube at 4000 rpm for 10 minutes. Aspirate the upper layer of urine into a new centrifuge tube, taking care not to aspirate exfoliated cells at the bottom of the urine in the tube.
2. Cell-free DNA extraction: The separated urine can be used for cell-free DNA extraction with a corresponding commercial kit.
